摘 要
桑黄是产多糖的药用真菌。本文以来自长白山的桑黄菌作为出发菌种,采用PDA培养基进行菌种活化,先测出桑黄的生长曲线和桑黄胞内、外多糖的积累曲线,然后用单因素实验法分别对桑黄液体培养基的初始pH值、碳源以及氮源进行了研究。结果表明:优化的碳源为玉米粉,氮源为黄豆饼粉,初始pH为5.5~6.5。优化培养基的具体配方为:玉米粉3%、黄豆饼粉1%、KH2PO40.3%、MgSO40.15%、维生素B120μg/100mL、维生素B230μg/100mL、pH在5.5~6.5之间。经过验证实验,在温度为30℃、130r/min的条件下培养桑黄,发酵7天后,优化培养基中发酵产生的桑黄菌丝体干重为7.089g/L,比PDA培养基中桑黄菌丝体产量提高了80.93%。
